Description

Hydroxychloroquine(HCQ) is a 4-aminoquinolone that is used to prevent and treat malaria, caused by parasitic protozoan from the Plasmodium genus, sold under the brand name “Plaquenil”. It is also used to reduce inflammation in the treatment of rheumatoid arthritis (see disease-modifying antirheumatic drugs) and lupus. Hydroxychloroquine differs from chloroquine by the presence of a hydroxyl group at the end of the side chain: the N-ethyl substituent is beta-hydroxylated. It is available for administration by mouth as hydroxychloroquine sulfate. Hydroxychloroquine(HCQ) is also shown to act as an antagonist for Toll-like receptors (TLR-7 and TLR-9) in plasmacytoid dendritic cells (pDCs).

Hydroxychloroquine was approved for medical use in the United States in 1955. It is on the World Health Organization’s List of Essential Medicines, the safest and most effective medicines needed in a health system. In 2017, it was the 128th most commonly prescribed medication in the United States, with more than five million prescriptions. The speculative use of hydroxychloroquine for COVID‑19 threatens its availability for people with established indications.
